{"id":14928,"date":"2018-04-05T10:29:28","date_gmt":"2018-04-05T10:29:28","guid":{"rendered":"https:\/\/www.revoscience.com\/en\/?p=14928"},"modified":"2020-05-27T06:01:18","modified_gmt":"2020-05-27T06:01:18","slug":"hkust-discovers-new-antibiotics-resistance-mechanism-paving-the-way-for-design-of-antibiotics-against-superbug","status":"publish","type":"post","link":"https:\/\/www.revoscience.com\/en\/hkust-discovers-new-antibiotics-resistance-mechanism-paving-the-way-for-design-of-antibiotics-against-superbug\/","title":{"rendered":"HKUST Discovers New Antibiotics Resistance Mechanism, Paving the Way for Design of Antibiotics Against Superbug"},"content":{"rendered":"<p><span style=\"color: #000000\"><em><strong>Researchers at The Hong Kong University of Science and Technology (HKUST) have identified for the first time the culprit behind the causing of broad-spectrum bacterial resistance to peptide antibiotics.<\/strong><\/em><\/span><\/p>\n<figure id=\"attachment_14929\" aria-describedby=\"caption-attachment-14929\" style=\"width: 768px\" class=\"wp-caption alignnone\"><img loading=\"lazy\" decoding=\"async\" class=\"size-full wp-image-14929\" src=\"https:\/\/www.revoscience.com\/en\/wp-content\/uploads\/2018\/04\/5435.jpg\" alt=\"\" width=\"768\" height=\"512\" title=\"\" srcset=\"https:\/\/www.revoscience.com\/en\/wp-content\/uploads\/2018\/04\/5435.jpg 768w, https:\/\/www.revoscience.com\/en\/wp-content\/uploads\/2018\/04\/5435-300x200.jpg 300w\" sizes=\"auto, (max-width: 768px) 100vw, 768px\" \/><figcaption id=\"caption-attachment-14929\" class=\"wp-caption-text\">The research team led by Prof Qian Pei-Yuan (second left) discovered for the first time that \u201cD-stereospecific resistance peptidases\u201d could render peptide antibiotics useless.<\/figcaption><\/figure>\n<p><span style=\"color: #000000\">Researchers at The Hong Kong University of Science and Technology (HKUST) have identified for the first time the culprit behind the causing of broad-spectrum bacterial resistance to peptide antibiotics &#8211; widely perceived as the last-line of defense against antibiotic-resistant bacteria, opening a new direction to the design of new drugs in tackling superbugs.<\/span><\/p>\n<p><span style=\"color: #000000\">The discovery \u2013 made by Chair Professor from the Division of Life Science Prof Qian Pei-Yuan and his fellow researchers, came in the best moment as the team has also discovered that this enzyme, which they identified as the cause of broad-spectrum bacteria resistance against peptide antibiotics, was actually found in not just a few, but many different strains of bacteria, sounding an alarm against persistent improper use of antibiotics.\u00a0<\/span><\/p>\n<figure id=\"attachment_14930\" aria-describedby=\"caption-attachment-14930\" style=\"width: 300px\" class=\"wp-caption alignright\"><img loading=\"lazy\" decoding=\"async\" class=\"size-medium wp-image-14930\" src=\"https:\/\/www.revoscience.com\/en\/wp-content\/uploads\/2018\/04\/5436-300x200.jpg\" alt=\"\" width=\"300\" height=\"200\" title=\"\" srcset=\"https:\/\/www.revoscience.com\/en\/wp-content\/uploads\/2018\/04\/5436-300x200.jpg 300w, https:\/\/www.revoscience.com\/en\/wp-content\/uploads\/2018\/04\/5436.jpg 768w\" sizes=\"auto, (max-width: 300px) 100vw, 300px\" \/><figcaption id=\"caption-attachment-14930\" class=\"wp-caption-text\">The specimen held by Prof Qian is peptide antibiotics already broken down by DRPs (the dot without the ring around it).<\/figcaption><\/figure>\n<p><span style=\"color: #000000\">Peptide antibiotics &#8211; including vancomycin and polymyxin for respective treatment of Staphylococcus Aureus (CA-MRSA) Infection and Escherichia coil infection, are often used as the last resort due to their resilience to multidrug resistant bacteria. However, a few years ago scientists have started identifying a few types of peptide antibiotics which have developed symptoms of bacterial resistance, although the causes behind remained unknown. Now, following the analysis of over 6,000 bacteria genomes under repeated validation through gene editing, chemical and enzymatic analyses, Prof Qian\u2019s team eventually identified a family of D-stereospecific resistance peptidases (DRPs) as the source of the problem, and its magnitude extends well beyond a few strains of bacteria.\u00a0<\/span><\/p>\n<p><span style=\"color: #000000\">\u201cDRPs are phylogenetically widely distributed in nature, if they are transferred to opportunistic pathogens with human\u2019s increasing misuse and overuse of antibiotics, more and more peptide antibiotics would be rendered useless, leading to delay or even failure in treatment,\u201d said Prof Qian, also David von Hansemann Professor of Science as well as Acting Head of the Department of Ocean Science at HKUST.<\/span><\/p>\n<p><span style=\"color: #000000\">\u2018Misuse and overuse of antibiotics of humans will intensify the problem of antibiotic resistance of pathogens, that makes research on peptide antibiotics even more important,\u201d he added. \u201cDeepening our understanding of antibiotic resistance mechanisms to peptide antibiotics does not only serve as a wake-up call, but is also conducive to increasing our collective antibiotic arsenal.
